[EC 3.5.4.9] an enzyme activity of the hydrolase class that catalyzes the cleavage of 5,10-methenyltetrahydrofolate to 10-formyltetrahydrofolate, a step in the system of folate-mediated one-carbon transfer reactions. The enzyme activity is part of a trifunctional enzyme that also includes methylenetetrahydrofolate dehydrogenase (NADP+) and formatetetrahydrofolate ligase activities.
