heavy chain disease A disorder of immunoglobulin synthesis in which large quantities of abnormal heavy chains are excreted in the urine. The amino acid sequences of the n- (amino-) terminal regions of these chains are normal, but they have a deletion extending from part of the variable domain through the first domain of the constant region, so that they cannot form cross-links to the light chains. The defect arises through faulty coupling of the variable (v) and constant (c) region genes.

They are the larger of the two types of polypeptide chains responsible for the biological and immunological properties of the different immunoglobulins. They differ according to the class of ig from which they were isolated, contain 450 to 600 amino acid residues per chain, and have molecular weights of 51-72 kD. myoD <molecular biology> Stands for myogenic determination.
MyoD was described as a master regulatory gene for the determination of muscle cells, but muscle determination is now thought to involve a family of genes. It is normally only expressed in myoblasts and skeletal muscle cells, but if transfected into cells will convert many differentiated cells into muscle cells. The myoD gene codes for the myoD protein which then switches on the the transcription of many muscle specific genes. MyoD is a member of a family of genes that all code for nuclear proteins which have a basic DNA binding motif and a helix loop helix dimerisation domain (bHLH or mHLH proteins, b=basic, m=myogenic). They bind to a consensus sequence CANNTG and can form homodimers or heterodimers with other members of the bHLH superfamily.

myosin light chain kinase <enzyme> An enzyme that phosphorylates myosin light chains in the presence of ATP to yield myosin-light chain phosphate and ADP, and requires calcium and calmodulin.
The 20-kD light chain is phosphorylated more rapidly than any other acceptor, but light chains from other myosins and myosin itself can act as acceptors. The enzyme plays a central role in the regulation of smooth muscle contraction.
