| 설명 | 핵산의 일종으로 DNA라고도 한다. Deoxyribonucleotide의 중합체이며 유전자의 화학적 본체이다. RNA바이러스 이외의 모든 생물은 DNA를 유전자로 지니고 있다. 디옥시리보뉴클레오티드(deoxyribonucleotide)는 염기와 당(2'-deoxy-D-ribose)과 인산으로 이루어진다. 염기는 아데닌(adenine), 구아닌(guanine), 티민(thymine)및 시토신(cytosine)의 4가지이며, 이것은 당에 부착되어 있다. 인산 역시 당의 한 부분에 부착되어 있다. 이 deoxyribonucleotide의 당은 다른 deoxy- ribonucleotide의 당과 인산을 사이에 놓고 결합을 하게 되어 하나의 긴 사슬을 형성하게 된다. 즉 당과 인산이 주축이 되어서 deoxyribonucleotide의 긴 사슬을 만든다. 이 deoxyribonucleotide의 사슬 두 개는 각각 deoxyribonucleotide에 부착되어 있는 염기들이 결합을 하여 두 개의 사슬이 결합되어 있는 이중나선 구조를 만들게 된다. 4가지 염기 아데닌은 티민과 결합을 하고, 시토신과 결합을 하게 된다. 즉 당과 인산은 긴 사슬을 만드는 역할을 하고 긴 사슬에 부착된 염기들의 결합에 의해서 두 개의 긴 사슬은 서로 붙어서 이중나선 구조를 만든다. DNA의 유전정보는 염기에 저장된다. 4개의 염기의 조합과 배열이 유전정보를 보관하는 하나의 암호 역할을 행하게 된다. |

| maxam-gilbert sequencing | A lab technique used to find out the sequence of nucleotide bases in a nucleic acid (a DNA or RNA molecule). The technique involves putting copies of the nucleic acid into separate test tubes, each of which contains a chemical that will cleave the molecule at a different base (either adenine, guanine, cytosine, or thymine or uracil (the last depending on whether it is DNA or RNA)). The result is that each of the test tubes contains fragments of the nucleic acid that all end at the same base, but at different points on the molecule where the base occurs. The contents of the test tubes are then separated by size with gel electrophoresis (one gel well per test tube, four total wells), the smallest fragments will travel the farthest and the largest will travel the least far from the well. The sequence can then be determined from the picture of the finished gel by noting the sequence of the marks on the gel and from which well they came from. (09 Oct 1997) |

| dideoxy sequencing | The most popular method of DNA sequence determination (c.f. Maxam Gilbert sequencing). Starting with single stranded template DNA, a short complementary primer is annealed and extended by a DNA polymerase. The reaction is split into 4 tubes (called A, C, G or T) each containing a low concentration of the indicated dideoxy nucleotide, in addition to the normal deoxynucleotides. Dideoxynucleotides, once incorporated, block further chain extension and so each tube accumulates a mixture of chains of lengths determined by the template sequence.
