GTP cyclohydrolase I [EC 3.5.4.16] an enzyme of the hydrolase class that catalyzes the first step in the biosynthesis of tetrahydrobiopterin from guanosine triphosphate. Deficiency of the enzyme, an autosomal recessive trait, causes malignant hyperphenylalaninemia.
