a fundamental equation of enzyme kinetics: v = Vmax[S]KM + [S] , where v is the “initial velocity” of an enzyme-catalyzed reaction (the velocity when the product concentration is near zero); [S] is the substrate concentration; and Vmax and KM are two constants that characterize a specific enzyme: Vmax, the maximum velocity, is the initial velocity seen when the enzyme is completely saturated with substrate, and KM is the Michaelis constant, defined operationally as the substrate concentration at which v = Vmax/2. The Michaelis-Menten equation does not apply to allosteric enzymes for which the binding of the substrate at the active site is altered by binding of the substrate at a second (allosteric) site.
