(for rickettsiae in smears), a thin smear is made in a phosphate buffer (pH 7.6) and air-dried, stained with methylene blue solution for 3 minutes, counterstained with safranine solution, and washed, blotted, and dried. Rickettsiae appear pale blue; cell nuclei and protoplasm are red.
