The disease is differentially characterised by high serum titres of antibodies to ribonuclease-sensitive extractable (saline soluble) nuclear antigen (nuclear ribonucleoprotein) and a speckled epidermal nuclear staining pattern on direct immunofluorescence. They do not solely affect connective tissues but they are autoimmune in origin.
