Kinins are cleaved from precursor substances called kininogens by kallikreins and are rapidly destroyed by kininases. Plasma kallikrein circulates as a proenzyme called prekallikrein and is converted to active form by factor XIIa, which cleaves HMW kininogen, an α2-globulin, to produce bradykinin.
