aflatoxin <chemical, microbiology, oncology> A fungal toxin which is a powerful liver carcinogen, found particularly in undeveloped and developing countries.
A group of closely related toxic metabolites that are designated mycotoxins. They are produced by aspergillus flavus and a. Parasiticus. Members of the group include aflatoxin b1, aflatoxin b2, aflatoxin g1, aflatoxin g2, aflatoxin m1, and aflatoxin m2.
Pharmacological action: carcinogens, mutagens, teratogens.
(27 Jun 1999)
aflatoxin b1 <chemical> A potent hepatotoxic and hepatocarcinogenic mycotoxin produced by the aspergillus flavus group of fungi. It is also mutagenic, teratogenic, and causes immunosuppression in animals.
It is found as a contaminant in peanuts, cotton seed meal, corn, and other grains. The mycotoxin requires epoxidation to aflatoxin b1 2,3-oxide for activation. Microsomal monooxygenases biotransform the toxin to the less toxic metabolites aflatoxin m1 and q1.
Pharmacological action: carcinogens, mutagens, teratogens.
Chemical name: Cyclopenta(c)furo(3',2':4,5)furo(2,3-h)(1)benzopyran-1,11-dione, 2,3,6a,9a-tetrahydro-4-methoxy-, (6aR-cis)-

aflatoxin m1 <chemical> Highly toxic 4-hydroxylated metabolite of aflatoxin b1 found in the milk of cows fed toxic meal.
It has mutagenic, carcinogenic, and teratogenic activity. It is formed by the enzymatic hydroxylation of aflatoxin b1 by aflatoxin b1 4-hydroxylase, a cytochrome p-450-dependent enzyme.
Pharmacological action: carcinogens, mutagens, teratogens.
Chemical name: Cyclopenta(c)furo(3',2':4,5)furo(2,3-h)(1)benzopyran-1,11-dione, 2,3,6a,9a-tetrahydro-9a-hydroxy-4-methoxy-, (6aR-cis)-

(12 Dec 1998)
aldose reductase A polyol dehydrogenase (NADP+);an oxidoreductase that reversibly converts aldoses to alditols (e.g., glucose to sorbitol) with NADPH as hydrogen donor. An important step in the metabolism of sorbitol and in the formation of diabetic cataracts.
See: d-sorbitol-6-phosphate dehydrogenase.
(05 Mar 2000)
aldose reductase inhibitor <pharmacology> A class of drugs being studied as a way to prevent eye and nerve damage in people with diabetes.
Aldose reductase is an enzyme that is normally present in the eye and in many other parts of the body. It helps change glucose (sugar) into a sugar alcohol called sorbitol. Too much sorbitol trapped in eye and nerve cells can damage these cells, leading to retinopathy and neuropathy.
Drugs that prevent or slow (inhibit) the action of aldose reductase are being studied as a way to prevent or delay these complications of diabetes.
