intradermal injection of a quantity of diphtheria toxin equal to one-fiftieth of the guinea pig minimal lethal dose in one arm (the test site) and of an equal quantity of heat-inactivated diphtheria toxin in the other arm (the control site). A positive reaction, which consists of an area of redness that appears in 24 to 36 hours at the test site only and persists for 4 to 5 days, leaving an area of brownish pigmentation, indicates a lack of immunity to diphtheria. A pseudoreaction, which consists of an area of redness appearing at both sites and usually disappearing in 48 hours without residual pigmentation, or a negative reaction indicates immunity.
